Assay for misreading of the resume codon by tmRNA triplet mutants. (A) tmRNA mutants were altered to encode the ANDH6D peptide tag provided that the ribosome resumes translation in the -1 (top) or +1 (bottom) frames. The maltose-binding protein with a stalling signal at the C-terminus served as a substrate for tagging, which was detected by anti-His6 antibodies. Addition of a known standard (tmRNA tagging in the 0 frame) at a 1:5 dilution allows comparison with the natural, 0 frame tagging process. (B) Quantification of tagging in the -1 frame relative to GUC (wild-type). Error bars represent the standard deviation of three independent experiments.
