Notch1 intracellular domain downregulates Snail protein. (A) Hep3B cells were transfected with Myc-Notch1 intracellular domain (NICD) and/or hemagglutinin (HA)-Snail and evaluated for the expression of both proteins using immunoblot analysis. (B) Hep3B cells were transfected with Myc-NICD and/or HA-Snail and evaluated for the expression of Snail mRNA using RT-PCR. (C) Hep3B cells were transfected with HA-Snail and/or different doses of Myc-NICD, then evaluated for the expression of both proteins using immunoblot analysis. β-actin served as an internal control. (D) Hep3B cells were transfected with HA-Snail and/or different doses of Myc-NICD and evaluated for the expression of Snail mRNA using RT-PCR. β-actin served as an internal control. (E) Hep3B cells were transfected with Myc-NICD and/or treated with 300 μM H2O2 for 72 hours, then evaluated for the expression of NICD and Snail proteins using immunoblot analysis.
