A working model of risiRNA biogenesis in C. elegans. The erroneous cellular rRNAs are scrutinized and suppressed through a number of mechanisms. Erroneous rRNAs are uridylated by CDE-1 and degraded by exoribonucleases such as SUSI-1(ceDis3L2). The disruption of CDE-1 or SUSI-1(ceDis3L2) results in the accumulation of erroneous rRNAs that thereafter recruit RdRPs to synthesize risiRNA. A risiRNA-mediated feedback loop silences rRNA expression through RNAi machinery and compensates for the disruption of the degradation of erroneous rRNA transcripts
